Researchers at the University of California, San Diego School of Medicine have uncovered a new signal transduction pathway specifically devoted to the regulation of alternative RNA splicing, a process that allows a single gene to produce or code multiple types of protein variants. The discovery, published in the June 27, 2012 issue of Molecular Cell, suggests the new pathway might be a fruitful target for new cancer drugs.

Signal transduction in the cell involves kinases and phosphatases, enzymes that transfer or remove phosphates in protein molecules in a cascade or pathway. SRPK kinases, first described by Xiang-Dong Fu, PhD, professor of cellular and molecular medicine at UC San Diego in 1994, are involved in controlling the activities of splicing regulators in mammalian cells.

Prior studies have implicated SRPK1 in cancer and other human diseases. For example, it has been shown that SRPK1 plays a critical role in regulating the function of Vascular Endothelial Growth Factor or VEGF, which stimulates blood vessel growth in cancer. SRPK1 has been found to be dysregulated in a number of cancers, from kidney and breast to lung and pancreatic.

Conversely, studies suggest the absence of SRPK1 may be problematic as well, at least in terms of controlling some specific cancer phenotypes. Reduced SRPK1, for example, has been linked to drug resistance, a major problem in chemotherapy of cancer.

In their new paper, Fu and colleagues place SRPK1 in a major signal transduction pathway in the cell. "The kinase sits right in the middle of the PI3K-Akt pathway to specifically relay the growth signal to regulate alternative splicing in the nucleus," said Fu. "It's a new signaling branch that has previously escaped detection."

As such, the SRPK offers a new target for disease intervention and treatment, researchers say. "It's a good target because of its central role and because it can be manipulated with compounds that suppress its activity, which appears quite effective in suppressing blood vessel formation in cancer," Fu said.

Alien world looms large in its neighbor's sky

Few nighttime sights offer more drama than the full Moon rising over the horizon. Now imagine that instead of the Moon, a gas giant planet spanning three times more sky loomed over the molten landscape of a lava world. This alien vista exists in the newly discovered two-planet system of Kepler-36.

"These two worlds are having close encounters," said Josh Carter, a Hubble Fellow at the Harvard-Smithsonian Center for Astrophysics (CfA).

"They are the closest to each other of any planetary system we've found," added co-author Eric Agol of the University of Washington.

Carter, Agol and their colleagues report their discovery in the June 21st Science Express.

They spotted the planets in data from NASA's Kepler satellite, which can detect a planet when it passes in front of, and briefly reduces the light coming from, its parent star.

The newfound system contains two planets circling a subgiant star much like the Sun except several billion years older. The inner world, Kepler-36b, is a rocky planet 1.5 times the size of Earth and weighing 4.5 times as much. It orbits about every 14 days at an average distance of less than 11 million miles.

The outer world, Kepler-36c, is a gaseous planet 3.7 times the size of Earth and weighing 8 times as much. This "hot Neptune" orbits once each 16 days at a distance of 12 million miles.

The two planets experience a conjunction every 97 days on average. At that time, they are separated by less than 5 Earth-Moon distances. Since Kepler-36c is much larger than the Moon, it presents a spectacular view in its neighbor's sky. (Coincidentally, the smaller Kepler-36b would appear about the size of the Moon when viewed from Kepler-36c.) Such close approaches stir up tremendous gravitational tides that squeeze and stretch both planets.

Researchers are struggling to understand how these two very different worlds ended up in such close orbits. Within our solar system, rocky planets reside close to the Sun while the gas giants remain distant.

Although Kepler-36 is the first planetary system found to experience such close encounters, it undoubtedly won't be the last.

"We're wondering how many more like this are out there," said Agol.

"We found this one on a first quick look," added Carter. "We're now combing through the Kepler data to try to locate more."

This result was made possible with asteroseismology. Asteroseismology is the study of stars by observing their natural oscillations. Sunlike stars resonate like musical instruments, due to sound waves trapped in their interiors. And just like a musical instrument, the larger the star, the "deeper" are its resonances. This trapped sound makes the stars gently breathe in and out, or oscillate.

Co-author Bill Chaplin (University of Birmingham, UK) noted, "Kepler-36 shows beautiful oscillations. By measuring the oscillations we were able to measure the size, mass and age of the star to exquisite precision."

He added, "Without asteroseismology, it would not have been possible to place such tight constraints on the properties of the planets."

Drug discovery and development company e-Therapeutics () is to start a US phase I clinical trial for its brain cancer drug ETS2101.

The trial is one of two studies scheduled for the drug, with a larger, UK-based trial studying the drug in patients with a variety of tumour types also starting shortly.

The US phase I trial is enrolling up to 24 brain cancer patients with primary brain cancer, called glioma, or cancer that has spread to the brain from other sites. First findings are expected in late 2012.

ETS2101 works by overcoming cancer cells? ability to evade apoptosis, which is the mechanism where cells normally self-destruct if they become dysfunctional.

It said it decided to also run a brain cancer trial because ETS2101 has shown it can cross between the circulating blood and the central nervous system fluid in the brain, unlike many current cancer drugs.

The primary objective of the trial is to evaluate safety and to determine an appropriate dose for phase II studies.

Development director, Steve Self, said: ?Advancing ETS2101 into trials was one of our key goals following last year?s refinancing and refocusing of our business.We look forward with interest to seeing the first results from the clinic.?

e-Therapeutics currently has three clinical candidates, spanning cancer, infectious diseases, such as hospital infection bug and MRSA, and psychiatric disorders.

e-Therapeutics uses its network pharmacology platform for drug discovery, which looks at all the effects a molecule might have on all the networks in the cell, unlike other methods which just target single proteins.

The debate can now be laid to rest: devices currently running on Windows Phone 7.5 will not be on the receiving end of the long-awaited Apollo update (aka Windows Phone 8). But that doesn't mean those smartphones are at the end of the line. It's long been speculated that the reason Microsoft would want to restrict WP8 to new phones is primarily due to improvements in hardware support and the change in platform architecture, and this conjecture appears to be right on the money. After all, how can single-core devices with WVGA screens take advantage of multi-core and additional resolution support? That part makes sense, but that leaves the software side.

So here's the plan to ensure current Windows Phone users stay happy: a few software benefits will be included in WP8, and a hearty helping of these killer features (such as the new Start screen) will be available to legacy devices in the form of Windows Phone 7.8. There'll be support for three tile sizes, with the smallest being fingertip size (a quarter of the standard square). The update will be delivered directly to users, sidestepping carriers, and can be done over WiFi.

Microsoft enters the post-PC era with the Surface tablet

Microsoft unveiled its tablet computer, Surface, on June 18. ?

Microsoft Corp. has swaggered into the post-PC era with the introduction of the Surface ? a nifty hybrid that is part tablet computer and part ultrabook.

It's a device with ambition ? hoping to take on the ubiquitous iPad, while also facing off against the multitude of MacBook Air PC clones.

Microsoft (NASDAQ: MSFT) made it clear the 10.6" Surface ? with its full-sized keyboards and multi-tasking capabilities ? is a full-blown PC in iPad clothing.

The kickstand-sporting Surface comes in two flavors. The Windows RT version will run on an ARM-based processors, while the Windows Pro version will have Intel's beefier Ivy Bridge processor.

Both models will run Microsoft's new operating system, Windows 8, and weigh just shy of 25 ounces. Pricing and availability are unknown.

The Surface shows Microsoft has realized what Apple always knew ? controlling the hardware and software means having to make fewer compromises on either.

With its hardware strategy, Microsoft has flipped the proverbial bird to its partners ? the hardware makers who license Windows operating system on their laptops and desktops.

For Hewlett-Packard, Samsung and Acer, Microsoft just went from full-time partner, to part-time competitor.

By going all in, Microsoft risks alienating some of its hardware partners.

Doing nothing, however, carries an even greater risk ? letting Cupertino not just define the post-PC era, but also own it.

'Catching Fire' Casting Rumors: Hunter Parrish Weighs In

'Weeds' actor, who was up for the role of Peeta in 'Hunger Games,' tells MTV News he'd 'love' to play Finnick in the sequel.

Our fellow "Hunger Games" fans will likely recall the fact that the casting process for the first film garnered more than a few headlines, particularly regarding the roles of Katniss, Peeta and Gale. Now that those roles have long been filled and the movie has gone on to break box-office records, we have been focused on the casting rumors around its sequel, "Catching Fire."

While Lionsgate hasn't made any announcements about who will join the cast for the sequel, particularly as fan-favorite character Finnick Odair, that hasn't stopped the rumor mill from churning out likely prospects. One of the fan-supported candidates is "Weeds" actor Hunter Parrish, who was also an early favorite to play Peeta until Josh Hutcherson won the role.

When MTV News caught up with Parrish recently to discuss the release of his first single, "Sitting at Home," we couldn't help but ask for his thoughts on the franchise and also being considered for Finnick.

"This whole 'Hunger Games' thing, man ... I've read all of the books, because I've been through this once before," Parrish said. "It was very exciting for me [to be considered] for the first movie, to be a part of that, and I read all of the books at that time, and I loved the script and the director and Jennifer Lawrence who was cast at that point. I saw it as something very exciting, and I was very excited about it, but it wasn't [meant to be]. Josh is also a friend of mine, and I haven't seen the film yet, but people say amazing things about it, and he's great, he's perfect. We've worked together before, and he's a great actor," Parrish said sincerely.

While Parrish is grateful for the fan support behind the Finnick campaign, his experience from the first round of casting has him feeling more cautious about things this time around.

"I kind of dove in for that one, so for this one, I don't know," he said. "I think you just kind of step back, and if it happens, it happens. I'm not going to really spend too much time thinking about it, but I'd love to. I wouldn't cast myself, but that's not helping me getting cast for that movie," he joked. "But I'm a fan of the books. I think there's fantastic actors that would probably be better. So there's that for getting me a job!"

In continuing with his gracious thoughts about other actors he'd consider for Finnick, we asked Parrish if he wanted to give a shout-out to any for fun.

"No, I wouldn't go that far!" he laughed. "I still want the job, I'm just saying ... "

Wall Street Week Ahead: Greek election results may bring respite

NEW YORK (Reuters) - Greek political parties supporting a bailout for the country won a slim parliamentary majority in Sunday's elections, which may give markets some respite, but any coalition's majority looks set to be narrow and may lack the stability needed to push through painful reforms.

Whatever the outcome, Europe's problems are far from over as the debt crisis threatens to further engulf the larger economies of Spain and Italy.

In a sign of relief over the Greek results, U.S. stock index futures opened up on Sunday, looking to extend the market's recent gains.

S&P 500 futures rose 6.2 points and were above fair value, a formula that evaluates pricing by taking into account interest rates, dividends and time to expiration on the contract. Dow Jones industrial average futures added 44 points and Nasdaq 100 futures rose 10.5 points.

"The gain is understandable given the massive fallout that could have happened had the (pro-bailout) New Democracy Party lost," said Michael Yoshikami, chief executive officer at Destination Wealth Management in Walnut Creek, California. "If they're able to construct a majority, as it appears, that will be a huge relief for the market."

The Greek conservative New Democracy party and Socialist PASOK, who broadly back an EU/IMF bailout package keeping Greece from bankruptcy, looked set to jointly secure a slim majority. SYRIZA, the leading leftist party that pledged to tear up the terms of the bailout package, conceded defeat.

If there is any sign of market stress on Monday morning, investors will look for action from the world's central banks who, according to officials, stand ready to intervene if trading becomes turbulent.

"We'll have all kinds of other headlines for Europe over the summer that we'll be trading on, so we're nowhere near the end of volatility," Yoshikami said.

At stake in Greece's election, as investors see it, had been the country's future in the euro zone and possibly the future of the currency bloc itself.

Official results released by the interior ministry, with 97 percent of ballots counted, showed New Democracy taking 29.7 percent of the vote and SYRIZA 26.9 percent. The PASOK Socialists were set to take 12.3 percent of the vote.

Because of a 50-seat bonus given to the party which comes first, that would give New Democracy and PASOK 162 seats in the 300-seat parliament.

The euro hit a three-week high against the U.S. dollar in early Australasian trade according to Reuters data, rising to around $1.2730 from around $1.2655 late in New York on Friday.

But markets have had a tendency to react positively to political developments late Sunday and early Monday only to quickly reverse. That was the case last weekend after the EU announced a 100 billion euro bailout for Spanish banks.

Weeks of worry over the potential outcome of the Greek election have prompted a number of central banks to prepare for market problems.

Central banks from major economies are ready to take steps to calm markets should the outcome of the Greek elections create a market storm, officials from the Group of 20 told Reuters.

Among them, European Central Bank President Mario Draghi said the ECB was ready to step in and fund any viable euro zone bank that gets in trouble. The Bank of England on Thursday announced a $155 billion (100 billion pound) offer of loans to banks.

G20 leaders kick off a two-day summit in Mexico on Monday and the rest of the week is not likely to be any quieter.

The Federal Reserve is due to release a policy statement on Wednesday at the end of its two-day meeting, and the steady flow of sovereign debt warnings and downgrades is likely to continue.

In another sign of investor nervousness, the CBOE Volatility index <.vix>, Wall Street's fear gauge, was up for much of Friday even as stocks rose, although the VIX closed lower. Stocks and the VIX typically have an inverse relationship.

"People have been hedging their positions aggressively over the past two weeks heading into this weekend," said Alec Levine, a derivatives strategist at Newedge Group SA in New York.

"No matter what happens (this) week, we will return to a massive game of chicken between the newly elected Greek government, whoever that may be, and the EU, specifically Germany."

THE FED AHEAD

Despite the fears, stocks ended the week on a positive note, marking a second straight week of gains. The benchmark Standard & Poor's index <.spx> is now up 6.8 percent for 2012, though still well off its highest levels of the year.

Part of what has spurred optimism for stock investors in recent weeks has been the hope that the Fed and other central banks would provide more economic stimulus. There has been continuing speculation over whether the Fed will engage in a third round of quantitative easing.

"We do think that expectations of QE3 will drive the market one way or the other," said Omar Aguilar, chief investment officer for equities at Charles Schwab Corp, in San Francisco.

But the fact that the Fed has made no recent changes to policy could mean the economic data policymakers are seeing is "not as bad as everyone thinks," Aguilar said.

Also ahead of the vote, Russell Indexes said certain events in Greece could mean changes in its indexes through implementation of its "financial crisis" rule. Its indexes include the Russell Global Index.

ON RATINGS WATCH

Adding to investor nervousness has been a slew of recent ratings cuts.

Among the most recent, Fitch Ratings on Friday downgraded Egypt's sovereign credit rating deeper into junk status. On Thursday, Egan-Jones cut France's sovereign credit rating.

Many investors see that trend continuing as agencies try to gauge the impact of the euro zone and other problems on the global economy.

"We're probably going to see more of it," Peterson said.

Wild almond tree oil may combat obesity, diabetes

ScienceDaily (June 18, 2012) ? A future weapon in the battle against obesity and diabetes could come in the form of an oil derived from the seeds of wild almond trees, according to researchers at Missouri University of Science and Technology.

The key to the oil's potential lies in its ability to affect certain microorganisms living in our bellies.

In a study presented June 18 at the American Society for Microbiology's general meeting in San Francisco, Missouri S&T researchers reported that adding sterculic oil to the diets of obese laboratory mice increased their sensitivity to insulin. This was due to the oil's effect on three types of microorganisms that live in the guts of the mice.

As a result, the researchers saw a "statistically significant improvement in glucose tolerance and insulin sensitivity in the obese mice," says Shreya Ghosh, a Ph.D. student in environmental engineering at Missouri S&T. The sterculic oil had no adverse affects on lean mice fed the same diet.

Sterculic oil is extracted from the seeds of the wild almond tree known as Sterculia foetida.

The research by Ghosh and her advisor, Dr. Daniel Oerther, builds upon previous studies conducted at the University of Missouri-Columbia. In those studies, sterculic oil was found to suppress the bodily enzyme stearoyl-CoA desaturase 1 (SCD1). SCD1 is associated with insulin resistance, a condition that can lead to diabetes and obesity.

Other studies have shown that obese mice deficient in the hormone leptin have a different composition of "gut microbiota" than do lean mice. (Those studies are referenced in a 2011 article in Nature Reviews Microbiology) Leptin helps regulate metabolism, and a deficiency of the hormone can contribute to obesity, says Oerther, the John and Susan Mathes Chair of Environmental Engineering at Missouri S&T.

In the Missouri S&T study, a diet supplemented by sterculic oil also correlated with lower levels of three types of gut microbiota -- Actinobacteria, Bacilli and Erysipelotrichia -- in the obese mice. It isn't clear, however, whether the lower levels of those microbiota led to the improvement of glucose tolerance and insulin sensitivity among the obese mice, Oerther says.

To perform her experiments, Ghosh studied 28 male mice -- 14 of them obese and 14 normal, and each of them five weeks old at the beginning of the study. She separated the mice into four groups and for nine weeks, fed a standard diet to one group of obese mice and one group of non-obese mice. Over the same period, she fed the same diet, supplemented with 0.5 percent of sterculic oil, to one group of obese mice and one group of non-obese mice. Ghosh recorded the weights, food consumption and glucose levels of the mice during the nine-week period.

After the nine weeks, researchers conducted a DNA analysis of the gut microbiota at King Abdullah Institute of Science and Technology in Saudi Arabia. The results confirmed correlations between the diet, improved glucose tolerance and groups of microbes. Even though the mice fed a diet with sterculic oil did not experience weight loss, both Ghosh and Oerther believe their findings could lead to new insights into controlling diabetes and weight gain.

Ghosh's research poster presentation at the ASM meeting is titled "Responses of Gut Microbiota to Sterculic Oil Supplemented Diet in Lean and Obese Mice." Her co-authors were Oerther; Dr. James W. Perfield II, assistant professor of food science at the University of Missouri-Columbia; and Dr. Pascal Saikaly, assistant professor of environmental science and engineering at King Abdullah University of Science and Technology in Saudi Arabia.
